The problems you have encountered with the low draw notes are very common. Holes 1 to 3, but especially hole 2, are dificult to get right. The problem is almost certainly your embouchure. It takes much practice to master.

: Finally, I play tuba as my primary instrument, would playing the harmonica effect my hombesure(sic) for tuba? I'm a high level on tuba and don't want my playing to suffer from playing harmonica.

I don't think it would make a difference. I don't play tuba, though, so I really can't say.
