First off welcome to the world of harmonica. You should start with the genre of music youre interested in playing IMHO. The key points to reading tab would be to understand what the tab is saying, inhale, exhale, bend, ect. This web site will be a big help. Start with the FAQs: General Questions. Reading thru this will be a good start. It explains the arrow system of tab. There are several different ways tab is written, and most are easy to understand. Now about that E harp, for playing cross harp, aka Blues harp, aka second position, you would be playing in the key of B. Now if you are playing straight harp, Key of E on youre E harp this would be the songs like "Oh Susana" ect. you just follow the tab. The key of B and B flat is popular with horn players. I would recommend starting with a Key of C harp to play in G crossharp. Many beginners books are for key of C harps. I would highly recommend getting a book with a CD or tape so you can hear the sounds youre after. Several very good choices are listed on this site. I hope this post helps. Good luck and feel free to e-mail me any questions.
