I don't think it matters if you're R or L handed. It matters where you are comfortable with having the low notes. I prefer them on the L side of my mouth. Most folks play this. There was one person I knew of that played with low notes on the R side of his mouth. Seems like Jimmy Reed but I don't think it was him. Play it how you like it. Otherwise you won't play it. That goes for what you play also.
